Aficamten

Aficamten Impurities

Aficamten is a next-generation cardiac myosin inhibitor under development for the treatment of hypertrophic cardiomyopathy (HCM). Its impurity profile includes structurally related analogs, residual solvents, and degradation products formed under stress conditions such as hydrolysis, oxidation, and photolysis. Furthermore, typical degradation pathways involve oxidative modification of the heteroaromatic scaffold, hydrolytic cleavage of amide bonds, and rearrangement of substituents within the carbamate or urea moieties. These impurities may affect the drug’s potency, stability, and safety. Importantly, all impurities are structurally characterized and qualified according to ICH guidelines for toxicological assessment and analytical validation.
